    • Role Description Department

    Applications are invited for a Postdoctoral Researcher to join the group of Professor Fred Antson in the Department of Chemistry at the University of York. This position is funded by a Wellcome Trust grant and is available for six months to study the structure and mechanism of rho helicase from Haemophilus influenzae. Based in the internationally renowned York Structural Biology Laboratory (YSBL), you will investigate the mechanism of nucleic acid unwinding by the helicase protein machinery. The research will necessitate protein production and purification and structure determination by cryo-electron microscopy and single particle 3D-reconstruction. The postholder will produce stable protein-nucleic complexes, suitable for structural biology. Structural studies, carried by using cryo-EM, will be complemented by biochemical characterisation with possible mechanisms of nucleic acid unwinding probed with suitable biochemical assays.

    Role

    To conduct research, in particular to:

  • produce Haemophilus influ rho helicase in E.coli and purify using chromatographic approaches
  • determine 3D structure by cryo-electron microscopy and single particle 3D-reconstruction
  • carry out complimentary biochemical and biophysical experiments
  • analyse possible mechanisms of the nucleic acid unwinding and probe these with suitable assays
    • Skills, Experience & Qualification needed
  • A first degree in Natural Sciences or related discipline
  • A PhD in structural biology or related discipline
  • Knowledge of molecular biology and genetics manipulation
  • Background in cryo-EM data collection and processing
  • Knowledge of a range of research techniques and methodologies
  • Skills in genetic engineering, molecular and structural biology
  • Highly developed communication skills and competency to conduct individual and collaborative research
  • Experience of writing up research work for publication
  • Experience in producing a helicase protein and in preparing and characterising a stable assembly with nucleic acids
  • Interest in and enthusiasm for research on molecular machines
  • Ability to develop research objectives, projects and proposals for own and joint research
  • Attention to detail and commitment to high quality
  • Positive attitude to colleagues and students
  • Commitment to personal development and updating knowledge and skills
