  • The European Training Network named ROPES (ROle of EPitranscriptomics in DiseasES) has been granted in the framework of the Horizon 2020 Marie Skłodowska-Curie Action for Innovative Training Network (ITN).

    The project is coordinated by the University of Trento (CIBIO) and it involves ten European laboratories and one biotech company from nine different countries, including: the Medical University of Wien (Austria), the University of Lausanne (Switzerland), the International Institute of Molecular and Cell Biology (Poland), the Umea University (Sweden), the German Cancer Research Center DKFZ (Germany), il Consiglio Nazionale delle Ricerche (Italy), the Centre for Genomic Regulation (Spain), CEITEC- University of Masaryk (Czech Republic), IMMAGINA Biotechnology Srl (Italy), Free University of Bruxelles (Belgium).

    The project aims to unravel the role of epitranscriptomics in diseases and to exploit it for novel therapies. ROPES, awarded with 3M € from the European Commission, started on December 2020, and will recruit Early Stage Researchers to perform innovative research projects in cooperation with leading universities in Europe and the United States of America. Twelve Early-Stage Researchers (ESRs) will be recruited to work in laboratories with expertise in RNA modification, development, bioinformatics, cancer, immunology, drug development.

    The objective of the ESR research project here advertised is to develop a virally-encoded tool to disrupt specific transcripts and trigger synthetic lethality in cancer cells.

    We expect to develop one or more approaches to trigger synthetic lethality in cells and mice. These approaches will provide the basis for a new line of treatment against cancer. The fellow will develop a non-mutagenic cytidine editing tool to generate C>U transitions and disrupt gene expression/translation. To this aim the fellow will optimize targeted editing tools in order to obtain high efficiency editing while avoiding the mutagenic effect of APOBEC1. Other available tools for targeted RNA editing will be tested for efficiency and off-target effects. The fellow will develop and use a computational approach to design a complete set of guide RNAs specific for genes in synthetically lethal genetic pairs. Induction of synthetic lethality will be then assessed using a panel of cell lines whose genetic milieu allows testing of distinct genetic pairs. The final test will happen in cancer-prone mice, to assess whether the RNA-targeted induction of synthetic lethality can be used to slow/stop cancer growth. The grant fellow is expected to apply and enroll in the Doctoral Programme in “Genetics, Oncology and Clinical Medicine (GenOMec)” at the University of Siena, Department of Life Sciences (academic year 2021/2022 - XXXVII cycle).

    Benefits

    CNR will stipulate a Research Fellowship (Assegno Grant) of 36 months within the duration of the ROPES project. The overall gross remuneration will include: - a living allowance of € 40.966,56 €/year - a mobility allowance of 7.200,00€/year - a family allowance (only if applicable) of 3.000,00€/year All amounts will be paid in monthly installments.

    The above-mentioned sums for Living/Mobility/Family allowances are to be considered gross amounts equal to the total cost that the employer must support (including taxes, social security and insurance that the Italian legislation places on CNR), with the sole exception of IRAP, the Regional Tax on Productive Activities borne by CNR. Both CNR and fellow's tax shares will be deducted at source.

    Candidates must demonstrate eligibility according to the Marie Sklodowska- Curie Actions eligibility requirements as follows:

  • Researcher status: Early-Stage Researchers (ESRs) are young researchers who, at the date of recruitment, are in the first four years (full-time equivalent research experience) of their research careers and have not been awarded a doctoral degree.
  • Nationality: Applicant ESRs can be of any nationality.
  • Mobility requirements: Applicant ESRs must not have resided or carried out their main activity (work, studies, etc.) in the country of the recruiting beneficiary for more than 12 months in the 3 years immediately prior to the date of recruitment

    The following skills/competences will be an advantage: • Background in either molecular and cell biology - in particular genome editing or computational biology.
