We require a dynamic, motivated person with a ‘can-do' attitude to address an exciting but challenging project funded by British Heart Foundation. The post will be located at the University of Nottingham (UK), and will collaborate with colleagues in King's College London (UK) and in the Cardiovascular Research Institute (Singapore).

The context is that hypertrophic cardiomyopathy (HCM) is a common genetic cause of heart malfunction, yet treatments are limited. In our past published work, we used CRISPR editing to introduce HCM mutations into β-myosin heavy chain (βMHC; MYH7) and cardiac actin (ACTC1) to create a suite of isogenic variants. Diverse molecular and functional phenotyping of the hiPSC-CMs, plus cross-referencing with patient-derived omics data and subsequent prioritisation, identified 15 coding & noncoding RNAs of new interest in HCM.

This project objectives are to:

1)) Create hiPSC-CM cultures in which novel RNAs are knocked out with CRISPR or upregulated by forced overexpression

2) Use high throughout/content and/or automated approaches to ask what impact modulation of RNAs has on gene expression, structure & function of hiPSC-CMs

3) Use a systems biology approach to integrate data on the transcriptome and proteome to unveil new mechanistic insight by identifying target pathways

The successful candidate will be nearing completion, or will have completed, a PhD in the biological sciences or equivalent. They will combine excellence in research, collaboration, communication and meeting deadlines with a solution- focussed attitude. The skillsets the candidate will have or will learn quickly include culture human pluripotent stem cells and direct their differentiation to cardiovascular lineages. Examples of existing or acquired skills are RNAseq, proteomics and bioinformatics; genetic engineering via CRISPR, RNAI and/or overexpression; immunostaining, flow cytometry, qPCR etc, supported by confocal microscopy and macro writing; formation of mono-cultures or co- cultures in 2D and 3D, including engineered heart tissue; DNA or RNA analyses or western blot; functional assays such as metabolism, cAMP, viability, electrophysiology, calcium flux and contractility.
