  • Context and Project Objectives: PROteolysis TArgeting Chimeras (PROTACs) are bifunctional molecules made up of a ligand for the target Protein Of Interest (POI) and a ligand for an E3 ubiquitin ligase: these ligands are covalently joined by a flexible linker. Their mode of action is the following: recruitment of the E3 ligase in close proximity to the POI, ubiquitination of the POI, recognition and degradation of the POI by the Ubiquitin Proteasome System (UPS). The goal of this project is the design, synthesis and biological evaluation of PROTACs targeting a protein overexpressed in most cancers. The intentional and controlled degradation of this protein will represent a therapeutical strategy to treat cancers.

    Job Description: This is a innovative and challenging interdisciplinary project, where the PhD student will be in charge of the synthetic part as well as the NMR conformational studies and biological evaluation of the final degraders. This is a great opportunity for the successful candidate to build a solid foundation in organic chemistry with applications to drug discovery and to acquire critical interdisciplinary skills in medicinal chemistry, chemical biology, biophysical methods for characterisation (NMR amongst others). In particular, the PhD student will carry out the synthesis of new ligands (small peptidomimetics), and incorporate them into PROTACs. In cells synthesis is also included in the scope of work. The PhD student will take advantage of a stimulating working environment within a young and dynamic team (with diverse / international backgrounds) with extensive experience in organic synthesis and biochemistry.

    Offer Requirements Specific Requirements

    Candidate's Requirements: The candidate must have a Master degree in Chemistry (or equivalent) and a very good theoretical and practical knowledge of organic synthesis as well as of the methods of analysis and characterisation of organic compounds. Experience in total synthesis and peptidomimetics / pseudopeptides synthesis will be a plus.
