A remarkable class of biopolymers known as ice-binding proteins alleviate the
risk of freeze injury throughout the Kingdoms of Life by keeping the
nucleation and growth of ice crystals in check. Yet, the application potential
of analogues of ice-binding proteins as cryoprotectants has remained
underexploited. This is because we are yet to unravel and utilize the
structure-function relations, which govern their activity as inhibitors of ice
recrystallization and promoters of ice nucleation at the single-molecule level
in vitro and within a complex biological environment. In this project you will
engineer new biosynthetic, ice-binding protein-polymers aiming to modulate
independently their ability to inhibit recrystallization, control nucleation,
and shape ice crystals. You will design and prepare various series of these
novel ice-binders to elucidate how to customize activity. You will study their
properties, assembly in solution, and activity. You will collaborate with
other group members to elucidate the mechanisms underpinning activity through
single molecule experiments of ice-binding as well as the application
potential in cardiac regenerative medicine.
PhD in biophysics. Experience with (computer-aided) protein engineering,
protein biophysics, and (polarized) optical microscopy; knowledge of
crystallization and microfluidics is preferred.
Ability to conduct high quality academic research, demonstrated by an
outstanding PhD thesis and publications in peer-reviewed journals.
Strong motivation to do excellent, original, fundamental research.
Excellent mastering of the English language, good communication and
leadership skills. Note that there is no Dutch language requirement.
Independent and goal-oriented scientist. Team player and able to work in a
dynamic, interdisciplinary, and international context.
