  • The thesis will take place in a Grenoble unit, the LPCV (Laboratory of Cell and Plant Physiology, CNRS, INRAE, CEA and University Grenoble Alpes Joint Research Unit). The LPCV has a recognized expertise in understanding the physiological mechanisms controlling the development and adaptation of plants to environmental constraints. These studies are performed at the molecular (genes, proteins), cellular (intracellular organelles) and whole plant levels. The LPCV is composed of about 120 people. It has plant culture facilities, and all the technical means to extract intracellular organelles (chloroplasts), purify proteins, characterize the physiological state of plants, analyze the dynamics of the subcellular localization of proteins (confocal imaging)... The thesis will be based on original data already available or being produced in the framework of an ongoing ANR project.

    The Chlorogenesis team of the Cell and Plant Physiology Laboratory (LPCV) is looking for a candidate for a thesis entitled "Control of chloroplast biogenesis". This thesis will be carried out within the framework of a project (C-TRAP) financed by the French National Research Agency (ANR) over the period 2022-2026. The candidate should have a strong background in biochemistry and genetics, with a certain degree of autonomy to manipulate proteomic and transcriptomic data. The candidate should have an interest in studies of higher plants and their ability to adapt to environmental constraints. He/she will develop his/her work in collaboration with the colleagues of the host team and in interaction with the partners of the ANR project. He/she will have to be both autonomous and have the ability to evolve in a multidisciplinary collective context.

    Description of the thesis topic: Preliminary data from the team have revealed unsuspected levels of post-transcriptional regulation highlighting important interactions between the translation of certain mRNAs encoded by the nuclear genome and the surface of chloroplasts. The identification of the targeted mRNAs allows to study the control of this association by environmental or developmental parameters. Access to the dynamics of the localization of these mRNAs during chloroplast biogenesis or plant responses to abiotic stresses is crucial to define the biological role and extent of these regulations. The thesis project proposes to address this fundamental question using several complementary approaches: subcellular fractionation and biochemistry of chloroplasts, classical molecular biology techniques, reverse genetics and plant phenotyping, but also state-of-the-art cell biology techniques, including real-time single-cell imaging of mRNAs in whole plants.

    This thesis is funded by the French National Research Agency (ANR) for the period 2022-2026 (C-TRAP project). The project involves two partners: the Cell and Plant Physiology Laboratory of Grenoble (CNRS, INRAE, CEA and University Grenoble Alpes Joint Research Unit), and the BIAM Laboratory of Cadarache (CNRS, CEA and University Aix-Marseille Joint Research Unit). Within the framework of this project, a PhD student will be recruited in Grenoble and a post-doctoral fellow will be recruited in Cadarache on the aspects targeting in particular the real-time single-cell imaging of mRNAs in whole plants. The selected student will thus take advantage of this multidisciplinary scientific context and will also benefit from a state-of- the-art technical environment, as well as from the access to various local or national experimental platforms. Finally, within the framework of CNRS, CEA, INRAE and the University of Grenoble Alpes, the student will also have access to various training courses.
