    Description of subject:

    In this project, we will study the ultrastructure and chirality of synthetic oligosaccharide crystals using two emerging technologies: Automated Glycan Assembly (AGA) at MPIKG and Cryo electron microscopy at Cermav. We will prepare cello- and chito-oligosaccharides with different chirality (i.e. different composition of D- or L-sugar units) with AGA. The structure of the resulting nanocrystals will be investigated using 3-D electron diffraction and tomography to understand how the molecular chirality affects the crystallization and morphology.

    Missions:

    Carbohydrate nanocrystals have shown potential for various applications. Their nanoscale chiral morphology, often observed as twists, can be exploited in optical materials and chiral catalysis. Such morphology is presumably related to the chirality of carbohydrate molecules. However, it remains elusive how the molecular chirality is transferred to the larger scale due to the lack of a model system suitable to investigate the crystallization behavior. Better understanding of the nanoscale chirality will allow us to develop strategies to rationally design carbohydrate-based chiral materials. In this project, we will study the ultrastructure and chirality of synthetic oligosaccharide crystals using two technologies: Automated Glycan Assembly (AGA) at MPIKG and Cryo electorn microscopy at Cermav. Synthetic oligosaccharides with different chirality (i.e. different composition of D- or L-sugar units) will be prepared with AGA. The structure and morphology of resulting chiral nanocrystals will be investigated using 3-D electron diffraction and electron tomography to understand how the molecular chirality affects the crystallization and morphology.

    Main activities:

    The successful candidate will work on both the synthesis and characterization of synthetic oligosaccharide nanocrystals. Their activities may include:

  • Synthesizing oligosaccharides with defined chirality with AGA.
  • Assembling synthetic oligosaccharides into nanocrystals.
  • Characterizing the resulting oligosaccharide nanocrystals using electron microscopy (3DED, tomography, etc).
  • Writing research articles and communicating in conferences based on the obtained results.
  • Structure description

    Centre de Recherches sur les Macromolécules Végétales (CERMAV) is a research center located in Grenoble, France and is dedicated to the science of plant macromolecules. Structure and properties of glycomaterials (SPG) team in Cermav, where Dr. Ogawa belongs, has a long-lasting expertise in the structural characterization of structural polysaccharides, such as cellulose and chitin.

    The Max Planck Institute for Colloids and Interfaces (MPIKG) is a multidisciplinary research institute located in Potsdam, Germany that combines chemical synthesis and materials science to bridge the gap between molecules and multiscale materials and biosystems. The carbohydrate materials group led by Dr. Delbianco uses automated technologies to prepare well-defined oligosaccharides as probes to study polysaccharides' conformation and aggregation.

    The project team is composed of two co-PI, Dr. Martina Delbianco at MPIKG and Dr. Yu Ogawa at CERMAV, and a PhD student.

    Previous formation, diplomas: Master's degree in chemistry, materials science or related fields.

    Expected background and skills: Since this project is interdisciplinary in the fields of organic and physical chemistry and materials science, the successful candidate may have a strong background either in (i) physical/analytical chemistry or (ii) organic chemistry. The knowledge and experience in electron microscopy and/or carbohydrate chemistry are beneficial but not necessary. Appropriate training will be provided on electron microscopy and organic chemistry.

    Personal skills:

    Ability to independently work in multidisciplinary research environment.

    Fluent in spoken and written English.

    Eligibility criteria

    Applicants must hold a Master's degree (or be about to earn one) or have a university degree equivalent to a European Master's (5-year duration).
