  • A 36 months PhD position in organic chemistry (starting date October 2021) is available in the group of Dr. Jean-Maurice Mallet at the laboratoire of Biomolecules (department of Chemistry, Ecole Normale Supérieure) to work on multidisciplinary project at the interface of chemistry and biophysics.


    Phagocytosis is a process that consists in the ability of a cell to internalize objects larger than 0.5 microns and it represents a fundamental part of innate immunity, organ homeostasis and tissue remodeling. This project aims at using micrometric oil in water (O/W) emulsion functionalized with fluorescent glycolipids in order to study phagocytosis induced by lectin receptors. Such lipid droplets can be fabricated in various sizes ranging from the nm to the µm scale, they are composed of natural triglycerides and are thus biocompatible and biodegradable and they display low immunogenicity unless properly functionalized with pathogenic patterns. Liquid particles have the advantage of allowing the mobility of the ligands at the interface upon interacting with a biological target. This mimics the clustering of ligands observed in vivo in cellular adhesion processes and increases affinity even with a low surface coverage of ligand. O/W emulsions are thus valuable tools to study adhesion and recognition processes like phagocytosis but to that end, they need to be functionalized with the proper ligands allowing for the specific recognition of cell membrane receptors and for their visualization by fluorescence microscopy.


    The PhD candidate will carry out the development of fluorescent glycolipids with which to functionalize the surface of the particles to provide them with the desired targeting and sensing abilities. The fluorophores will be designed either as FRET pairs to study the interactions between different membrane receptors involved in the cellular adhesion or as environmentally-sensitive probes to evidence changes in pH or redox processes during the degradation of the particle.

    This PhD thesis will take place in the frame of the PhagoChemiForce project funded by the Agence Nationale de la Recherche (ANR). It is a collaboration between three teams of chemists (Jean-Maurice Mallet, UMR 7203, Ecole Normale Supérieure), biophysicists (Jacques Fattaccioli, UMR8640, Institut Pierre Gilles de Gennes) and biologists (Florence Niedergang, Institut Cochin).

    The successful candidate should hold a master degree in molecular chemistry with a solid background in organic synthesis and be motivated to work on a multidisciplinary and collaborative project that will require him/her to learn new experimental skills. Good oral and written communication skills in English are required.
