  • The PhD program in Biomolecular Sciences is a research doctorate of the University of Campania Luigi Vanvitelli in Caserta-Naples, Italy. The program is multidisciplinary and based on a well-balanced combination of research activity with access to the most modern technological platforms, theoretical courses, internal seminars and participation to national and international conferences. The official language of the Doctorate program is English. Research and training activities will take place in the Department of Environmental, Biological and Pharmaceutical Sciences and Technologies of the University of Campania Luigi Vanvitelli, via Vivaldi, Caserta and in the CNR Institutes of Genetics and Biophysics A. Buzzati-Traverso, Institute of Biochemistry and Cell Biology, and Biostructures and Bioimaging of Napoli. All these institutions provide an international and very stimulating research environment for the students. The students will spend part of their doctorate (at least 3 months) in research groups abroad.

Your project: Transition metal ions are important cofactor for numerous biochemical reactions. They have been largely investigated for their main roles as structural and catalytic cofactors for biomolecules present in living systems as they are able to facilitate many cellular processes such as signaling, proliferation, DNA synthesis and repair. The molecular rules regulating metallo-proteins function are buried in the relationship between sequence and structure, and therefore require deep understanding of their folding mechanism, reactivity and metal-protein affinity. Yet, how metal- dependent proteins fold as well as the factors modulating metal-protein affinity and stability remains still poorly understood. Protein folding is totally metal-dependent in some instances, involving the early coordination of the metal cofactors that, in turn, address the folding into the functional native conformation. On the contrary, metal ions can bind proteins in a later stage of the folding pathway. The relationship of the metal cofactors, protein folding and structural stability can be properly characterized by an integrated approach in which spectroscopic and calorimetric data are combined together. The study of the influence that different metal ions, with their different electronic structures, have on protein folding/unfolding pathways will be the main goal of the proposed Ph.D. project, intended to contribute to the search for the roles of metal ions in protein folding reactions.
