  • In vitro generation of micro-tissues is becoming the alternative to animal testing and assessment of pathologies and treatments in personal medicine. Such models require perfused microvascularization to mimic tissue physiology and to allow delivery of drugs. Our project aims at combining cell self- assembly with microfabrication to generate micro-tissues with a perfused capillary network.

    We will develop innovative microfluidic devices consisting in two layers connected with fluidics, assembled with magnetic bonding. Both

    layers will contain channels coated with matrix proteins and seeded with cells. The central area of the bottom layer will contain a collagen

    hydrogel cellularized with endothelial cells co-cultured with stem cells to promote formation of a capillary network. Microfabricated channels with microvascular network in hydrogels will be connected through anastomosis resulting from promotion of angiogenesis by mesenchymal stem cells. This project will provide a tool for the generation of vascularized skin substitute with a wide range of applications from basic research to disease modelling and pharmacological testing.

    This project, funded by the ANR, is a collaboration between Carole Aimé, at the Chemistry Department at ENS, and Laurent Muller at the Center for Interdisciplinary Research in Biology, Collège de France.

    Benefits

    PerVaSkin combines the expertise of biologists in in vitro 3D angiogenesis and skin physiology with that of chemists and physicists in microfluidics, in order to address a major challenge in biotechnology: generating a perfused microvascularized skin substitute on a chip. Such device bio-mimicking skin at best will provide a unique model that is still a missing step in the spectrum of skin substitutes available. Beyond its use in basic and applied research, this skin-on-chip should provide a powerful link with the clinic by allowing to set up: -1/ preclinical evaluation of therapeutic drugs and -2/ relevant assays based on patient-derived skin cells.

    Eligibility criteria

    The PhD student will be involved during the entire project, from microfabrication of scaffolds for the “vascular chip”, to differentiation of keratinocytes in the skin-chip. This student should thus have good background in cell biology and a strong interest for the development of nano and micro technologies for biomedicine. The PhD student will work in close collaboration with Carole Aimé at ENS, and with Laurent Muller at the Collège de France.
