  • The candidate will work in Corinne Dupuy's group and will have (a) to participate in the design and implementation of experiments, (b) to manage the organization of these experiments, (c) to adapt experimental protocols, ( d) to autonomously carry out the quantitative analysis of the data including the appropriate statistical tests, (e) to present the results obtained during laboratory meetings, (f) to participate in the writing of manuscripts for the publication of the results, (g) to be able to supervise young researchers (M2/PhD)

    The objective of the project is to develop an innovative strategy targeting an H2O2-producing enzyme: NADPH oxidase DUOX1, which contributes to the development of pulmonary fibrosis (Louzada RA et al. Eur Respir J. 2021 57(1):1901949) . This therapeutic strategy is based on the use of aptamers, oligonucleotides that can fold into complex 3D structures and bind to targets with high affinity and specificity and thus, inhibit their function. We plan a) to identify and characterize, from a functional point of view, candidate aptamers by analyzing their ability to inhibit the process of myofibroblastic differentiation from murine and human pulmonary fibroblasts and, b) to evaluate the therapeutic efficacy of selected aptamers using a mouse model of bleomycin-induced pulmonary fibrosis. The experiments will be conducted in vitro on primary cultures of pulmonary fibroblasts and in vivo on mice and will use techniques for analyzing the expression and activity of proteins (measurement of H2O2) and mRNAs: immunofluorescence, cytometry, immunoprecipitation, western blot, qPCR, primary culture preparation

    Our group is part of the F. Rosselli's team in the UMR9019 CNRS "Genome Integrity & Cancers" research unit located at the Gustave Roussy Institute. The unit and the institute have the necessary equipment for the well running of the experiments. The candidate will benefit from the expertise of a scientific environment of the Unit and will participate in the development of a new project of the team

    Eligibility criteria

    The candidate must have experience in a research laboratory and great rigor in carrying out experiments. At the experimental level, advanced skills in cell biology (primary culture preparation, transfection), molecular/biochemical biology, fluorescence imaging and cytometry are required. At a fundamental level, a good knowledge of redox systems and the processes involved in the development of pulmonary fibrosis is also required.
