    Created on the initiative of researchers and physicians at the Necker Hospital - Enfants malades, the Imagine University Hospital Institute (IHU) develops scientific programs based on continuity between fundamental research, clinical research and innovative care for genetic diseases and genetic predispositions to frequent diseases, from childhood to adulthood. Imagine's ambition is to accelerate research by organizing it as close as possible to the patient in order to provide the diagnostic and therapeutic solutions so eagerly awaited by patients and their families.

    A postdoctoral Fellow position is available in the lab of Genome Dynamics in the Immune System (supervised by Erika Brunet).

    MAIN MISSIONS

    Project description: Targeting Anaplasic Large Cell Lymphoma cells with CART cells and Antibody Drug Conjugates

    Anaplastic large-cell lymphoma (ALCL) is an aggressive T lymphoma. Approximately 84% of ALCL cases harbor a fusion involving the anaplastic lymphoma kinase (ALK) gene and most often the NPM gene, with high prevalence in children. This results in a constitutively activated fusion protein NPM-ALK driving the disease by activating oncogenic signaling pathways. Most of ALK+ ALCL cases initially respond well to the currently applied frontline chemotherapy, but are prone to develop resistance with around 30% of patients relapsing; relapses that are associated with a poor prognosis. In a recent study (Babin et al, Mol Cancer 2022), we demonstrated high-efficacy transformation of primary human activated T lymphocytes upon precise genome engineering of the NPM-ALK translocation. Using this original model, we identified a cell surface receptor as a robust and genuine marker of all ALK+ ALCL patient tumor samples tested. The membrane localization of this receptor and its very restricted pattern of expression in adult tissue (it acts mostly in early embryogenesis with decreasing expression during ontogeny to almost undetectable levels in most adult organs) constitutes a very promising surface target for ALK+ ALCL. These characteristics open the road to direct targeting by 2 different strategies: (i) monoclonal antibodies conjugated with cytotoxic drugs (Antibody Drug Conjugate, ADC) (ii) specific CAR T cells.

    ACTIVITY

    Objective of the research: Selecting the most efficient therapeutic strategy (CART or ADC) to specifically kill ALCL ALK+ cells with perspective for preclinical studies

    This project will benefit from active collaborations with Dr T.Mercher (Insitut Gustave Roussy) and Dr F. Meggetto (CRCT, Toulouse) and is part of a collaborative PLBIO-INCA granted project.

    REQUIRED SKILLS

    The candidate should have strong skills in cell biology analysis (ex: multiparameter flow cytometry) and primary lymphocytes cultures. She (he) should be trained for basic molecular biology and biochemistry analyses, be self-motivated, well-organized and proactive with strong communication and technical skills. She (he) should be able to work independently, lead the project and to collaborate effectively with the team and external collaborators. English language should be fluent for reading, writing and discussing results.

    Preferred skills : Experience in immuno-oncology .

    QUALIFICATIONS

    A PhD/MD-PhD degree that includes a strong experience in oncology and/or Immunology.

  • Fixed-term contract funded for a 1 year with a possible 2-year extension
